Reverse Pharmacology in Ayurveda: Integrating Traditional Knowledge with Modern Drug Discovery

The complexity, cost and failure rates of conventional drug development have prompted researchers to explore alternative approaches for the identification of novel medicinal compounds. Reverse pharmacology is a new and effective strategy that starts with clinical observation and traditional medical knowledge and confirms the outcomes via scientific investigation. Ayurveda, one of the oldest healthcare systems in the world, is a large collection of documented clinical experiences and therapeutic practices, accumulated over thousands of years. Reverse Pharmacology uses this experience information to accelerate the drug discovery process, reducing the time, expense and risk involved in the conventional drug research and development. “This is a clinic-to-laboratory approach rather than the traditional laboratory-to-clinic approach.” The present review paper discusses the concept, development, scientific principles and three-stage model of reverse pharmacology and its application in Ayurveda. The article also presents successful examples, current drug development applications, difficulties, constraints and future prospects of artificial intelligence, omics technologies, systems biology, network pharmacology and precision Ayurveda. This confluence of old ayurvedic wisdom and contemporary scientific methodologies opens up great opportunities for the discovery of safe, efficient and inexpensive medicines for worldwide healthcare.
